Once you enter a birthday and click confirm, that is your permanent zodiac sign.

how come I killed zodiac eater 4 times and never got the rewards like shard or enchantment stone? are they that rare?

You have to have the quest active to get rewards from killing a zodiac boss. You can complete one quest per day, accepted in the zodiac panel.
